TranscriptomeanalysisofChouioiacuneaYanginresponsetoavermectinstressJINXin1,2,QINDongyu1,2,LIUXinyu1,2,ZHANGTiantian1,2,RENRui1,2,GUOMeiqi1,2,LIMin1,2,ZHUGengping1,2,PANLina1,2(1.CollegeofLifeSciences,TianjinNormalUniversity,Tianjin300387,China;2.TianjinKeyLaboratoryofConservationandUtilizationofAnimalDiversity,TianjinNormalUniversity,Tianjin300387,China)Abstract:ToexplorethestresseffectofpesticideswidelyappliedinagricultureandforestryonChouioiacuneaYang,thesafetyofavermectin,whichisakindofantibioticinsecticide,onC.cuneawasassessedbythemethodoffilmbioassay.TranscriptomesequencingwasusedtoanalyzethealternationofcertaingeneexpressionofC.cuneaunderavermectinstress.Theresultsshowedthat:①BecauseofthelowtoxicityofavermectinwiththeconcentrationusedinthefieldtoC.cunea,avermectinisconsideredtobealowriskpesticide,whilehighconcentrationstresshadshort-termkillingeffectonC.cunea;②Comparedwiththecontrolgroup,atotalof805differentiallyexpressedgeneswereobtainedbytranscriptomesequencingofC.cuneaunderavermectinstresswithLC10andLC30concentrations,ofwhich33geneswereup-regulatedand772genesweredown-regulated;③ThefunctionalanalysisofGeneOntology(GO)annotationrevealedthatthecorrespondingdifferentiallyexpressedgeneswereenrichedincellprocess,metabolicprocess,membraneandmembranecomponents,molecularbindingandcatalyticactivities;④TheanalysisofKEGGmetabolicpathwaydemonstratedthattheinteractionofneuroactiveligand-receptorsandthecytochromeP450-induceddrugmetabolizingpathwayweresignificantlyenriched;⑤DetectionofcytochromeP450-relatedgenesbyRT-qPCRshowedthattheexpressionlevelsofCcCYP9AG3andCcCYP9AG4weresignificantlyhigherintheavermectinstressgroupthaninthecontrolgroup,whichindicatedthatthecytochromeP450genefamily,inparticular,theCYP9familyisinvolvedintheresponseofC.cuneatoavermectinstress.Keywords:ChouioiacuneaYang;avermectin;toxicitydetermination;stressresponse;transcriptomesequencing周氏啮小蜂对阿维菌素胁迫响应的转录组分析金欣1,2,覃东玉...
