中国国境卫生检疫杂志2023年2月第46卷第1期ChineseFrontierHealthQuarantineFeb.2023,Vol46,No.1ComparisonofthestabilityofmatrixsolutionofHBsAgweaklypositivequalitycontrolserumLUQin,SUJinkun,CHENJia,LIXueming,QIPeiyan,MINGHongyanGuangzhouInternationalTravelHealthcareCenter,Guangzhou,Guangdong510635,ChinaAbstract:ObjectiveToselectdifferentmatricestopreparehepatitisBsurfaceantigen(HBsAg)indoorqualitycontrolserumforELISA,andtofindastablematrixsolutionforthepreparationofHBsAgweaklypositivequalitycontrolserum.MethodsAserummixtureofnegativeHBsAg,anti-HIV,anti-HCVandanti-treponemalwascollectedasnegativeserum(HBV-A),fourmatricesincludingnegativeHBVserum(HBsAg,HBsAb,HBeAg,HBeAg,HBeAb,HBcAb)andnegativeforanti-HIV,anti-HCV,anti-treponemalwerecollectedasantibody-negativeserum(HBV-B),aswellasfourmatrices,humanalbumin(HBV-C)andfetalbovineserum(HBV-D)wereused.Accordingto1∶1000,1∶2000,1∶4000,1∶8000,1∶16000,1:32000,1:64000dilutedHBsAgpositiveserum,adilutionseriesofdifferentmatriceswerepreparedtodetectHBsAgbyELISAmethod.2-4timesofthecut-offvaluewasconfirmedasthetargetS/COvalue.Thefourmatrixdilutionseriesstoredat4°Cfor1,3,5,7,9,11d,1monthand2months,targetS/COvaluecorrespondstothestabilityoftheconcentrationwerecompared.Thedilutionseriespreparedwiththreeselectedmorestablematriceswereobservedfor5weeksat4℃and6monthsat-20℃,andthecorrespondingconcentrationoftargetS/COvalueswascompared,andthestabilityofthequalitycontrolserumpreparedwiththethreeselectedmatriceswascomparedfor15monthsat-20℃.ResultsTheHBsAgdilutionseriespreparedwithHBV-Amatrixhadpoorstability,andtheconcentrationcorrespondingtothetargetS/COvaluewasreducedby10afterwhenstoredat4℃for1week.Dilutionseriespreparedassubstrateswereplacedat4℃for5weeksand-20℃for6months,withstableconcentrationscorrespondingtotargetS/COvalues.Thequalitycontrolserumpreparedbythethreeselectedmatriceswasplacedat-20℃for15months,andthestabilitydifferencewasnotsignificant(P>0.05),thestabilitywas...
