南开大学学报(自然科学版)ActaScientiarumNaturaliumUniversitatisNankaiensisVol.56№1Feb.2023第56卷第1期2023年2月ArticleID:0465-7942(2023)01-0046-08MCM6ContributestotheProliferationofEsophagealCarcinomaCellsandPredictsPoorPrognosisLiDemao1,SunHuijie2,MengLinglei3,LiDeshang4(1.DepartmentofThoracicSurgery,XingtaiPeople'sHospital,Xingtai054000,China.2.DepartmentofPharmacy,XingtaiMedicalCollege,Xingtai054000,China;3.DepartmentofCT/MR,XingtaiPeople'sHospital,Xingtai054000,China;4.DepartmentofLaboratory,XingtaiPeople'sHospital,Xingtai054000,China)Abstract:Esophagealcarcinoma(ESCC)isacommontypeoffatalcancerwithhighmortality.Thepurposeistoidentifykeygenesanddeterminethepotentialmechanismsinvolvedinthedevelopmentofesophagealcarcinomabasedonbioinformaticsanalysis.Chromosomemaintenance6complex(MCM6)hasbeenidentifiedtobeup-regulatedinavarietyofmalignancies.However,itseffectsonesophagealcarcinomaremainunclear.UsingtheGEPIAdatabase,itwasinvestigatedhowMCM6expressionvariedbetweenhealthyandcancerousesophagustissues.AndtheresultthatESCCwithhighMCM6geneexpressionwasfound.Thenimmuno-histochemistry(IHC)assaywasperformedtodetectMCM6expressioninbothhealthyandmalignantesophagealtissues,andtheresultshowedhighMCM6proteinexpressioninESCC.Theχ2analysistoexplorethecorrelationbetweenclinic-pathologicalfeaturesandproteinexpressionandthetumorsizeofesophagealcancerwassubstantiallyassociatedwithMCM6expressionin78patientsinourhospital.Byperformingacolonyformationassay,thepromotingeffectofMCM6ontheproliferationofesophagealcancercellswasidentified.TumorgrowthassayconfirmedthepromotingroleofMCM6intheprogressionofesophagealcancerinvivo.ThecorrelationbetweenMCM6andPCNAwasdetectedbydatabaseandimmunohistochemistry,andwefoundthatMCM6waspositivelycorrelatedwithPCNAexpression.Further,itwasspeculatedthatMCM6mightpromotetheproliferationofesophagealcancercellsbymediatingPNCAexpression.Inconclusion,MCM6mightbecomeanewtargetforthetreatmentofesophagealcarcinoma.Keywords:MCM6;PCNA;esophagealcarcinoma;proliferatio...
