Isolation,identificationanddegradationcharacteristicsoforganicmatterdegradingbacteriainblackandodorouswaterMAXing-guan,DONGChang,TANGYu-lan,HEYa-ting,WANGXiao-hua(SchoolofMunicipalandEnvironmentalEngineering,ShenyangJianzhuUniversity,Shenyang110168,China)Abstract:Thisworkaimedtoexcavatehigh-efficiencyorganicmatterdegradationstrainfromblackandodorouswater,whichwasappliedtolow-concentrationorganicpollutedblackandodorouswater,andevaluateitsabilityandcharacteristicstodegradeorganicmatterinblackandodorouswater.Blackandodorouswaterorganicmatterdegradingstrainwasobtainedbyroutineisolationandscreeningofbacteria,andthestrainwasidentifiedthroughmorphologicalobservationand16SrDNAgenesequence.Thesingle-testexperimentswereusedtooptimizecultivationconditions.Furthermore,undertheoptimizedcultivationconditions,thegrowthcurveofthestrainwasmeasuredtodeterminetheoptimalinoculationpoint.ThedegradationabilityofthestrainonorganicmatterinblackandodorouswaterandthedegradationcharacteristicsunderdifferentinitialbacterialconcentrationsandcarbonsourceswereevaluatedbymeasuringCODCrremovalrateandcolonynumber.Ahighdegradationrateoforganicmatterinblackandodorouswaterbacteriumawasisolatedandscreenedfromthesedimentofblackandodoroussection(RiverRenjing,Shenyang),whichwasidentifiedasBacillustoyonensis.TheoptimalgrowthconditionsafteroptimizationwerepH7.0,equipmenttheliquidvolumewas25%,thetemperaturewas32℃.Underthiscondition,theoptimalinoculationpointwasculturedfor12h.Theorganicmatterdegradationcharacteristicsofstrainashowedthattheremovalrateoforganicmatterinblackandodorouswaterreached64.75%whenthereactionwascarriedoutinashakerat30℃and120r/minfor4days.Underthesameexperimentalconditions,increasingtheinitialconcentrationofbacteriaresultedinincreasinginCODCrremovalrateandthendecreasing.Whentheinitialbacterialconcentrationwas1%(volumefraction),CODCrremovalratewashigher.Strainahaddifferentutilizationofdifferentcarbonsourcesandhadhigherutilizationofsmallmolecularweight.Th...
