Panax ginseng extract,UPGE)对慢性不可预知温和刺激(chronicunpredictable mild stress,CUMS)模型小鼠抑郁样行为及下丘脑-垂体-肾上腺(hypothalamic-pituitary-adrenal,HPA)轴的影响。方法:超高效液相色谱-串联质谱(UPLC-MS/MS)方法鉴别 UPGE 中的皂苷成分。将 60 只雄性昆明小鼠随机分为空白组(Control),模型组(CUMS),文拉法辛组(10 mgkg1 Ven)以及林下参提取物组(100 mgkg1 UPGE)。除空白组外,均采用 CUMS 诱导各组小鼠抑郁,灌胃给药 21 d 后通过小鼠体质量,进行糖水偏好实验,旷场实验,水迷宫实验分析其行为学变化;采用酶联免疫吸附测定法(enzyme linkedimmunosorbent assay,ELISA)检测促肾上腺皮质激素释放激素(corticotropin-releasing hormone,CRH),促肾上腺皮质激素(adrenocorticotropic hormone,ACTH)和皮质酮(corticosterone,CORT)的含量,苏木精-伊红染色(hematoxylin-eosin staining,HE)法观察小鼠海马 CA1,CA3 区神经元细胞病理变化,原位末端标记法(TUNEL)显色法检测海马区细胞凋亡情况。结果:UPGE 中含 16 种皂苷成分。相比于模型组,UPGE 给药组小鼠糖水偏好度显著提升(P0.01)、旷场实验中水平运动距离增加和垂直站立次数增加(P0.05,P0.01)、水迷宫实验中逃避潜伏期降低(P0.05)、增加目标象限停留时间、穿越平台次数(P0.05,P0.01)、能够缓解海马神经元细胞的损伤,血浆中 ACTH、CORT 和 CRH 水平降低(P0.05),海马区细胞凋亡率降低(P0.05,P0.01)。结论:UPGE 有显著的抗抑郁作用,其作用机制可能与调节 HPA 轴功能有关。关键词:林下参,慢性不可预知温和刺激,下丘脑-垂体-肾上腺轴,抑郁本文网刊:中图分类号:TS201.4 文献标识码:A changes.Thecontents of corticotropin-releasing hormone(CRH),adrenocorticotropic hormone(ACTH)and corticosterone(CORT)inplasma were measured by enzyme linked immunosorbent assay(ELISA).Hematoxylin-Eosin(HE)staining was used toinvestigate the pathological changes of neurons in the CA1 and CA3 area of hippocampus.The apoptosis of hippocampalneurons was observed by terminal-deoxynucleoitidyl transferase mediated nick end labeling(TUNEL)method.Results:UPGE contained 16 ginsenosides.Compared with the model group,the UPGE group significantly improved sugar-waterpreference(P0.01),increased horizontal movement distance,and increased vertical standing times in the absenteeismexperiment (P0.05,P0.01).Shortened the escape latency in Morris water maze test (P0.05).Increased the targetquadrant residence time and the platform crossing times in Morris water maze test(P0.05,P0.01).UPGE would alleviatehippocampal neuron injury.The levels of ACTH,CORT and CRH in plasma were decreased(P0.05),the apoptosis ofhippocampal cells was decreased(P0.05,P0.01).Conclusion:The UGPE would improve antidepressant behavior ofCUMS mice,and its mechanism might be 抑郁症是一种中枢系统精神障碍,以情绪低落,思维迟钝,语言行动减少,绝望等为主要表现,难治愈且易反复发作1。发病机制与下丘脑-垂体-肾上腺(hypothalamic-pituitary-adrenal,HPA)轴有关。HPA轴是脑内重要的神经内分泌反馈调节轴,在面对应激时,由于促肾上腺皮质激素释放激素(corticotropin-releasing hormone,CRH),促肾上腺皮质激素(adre-nocorticotropic hormone,ACTH)和皮质酮(cortico-sterone,CORT)的大量释放,HPA 轴过度激活,导致功能亢进,负反馈调节机制受损,并伴有海马神经元的损伤23。目前临床抑郁症治疗多采用神经递质再摄取抑制剂并辅以心理治疗,但疗效差并伴有副作用。中药治疗抑郁症不仅可以缓解症状,并可长期服用,具有多成分,多靶点的特点,能够成为抗抑郁药物重要来源4。林下参(understory Panax ginseng,UPG)是把人参种子撒播到山林后数年采收,主要成分为人参皂苷、多糖、挥发油等物质,具有调五脏,安精神,定魂魄之功效5。研究证实人参皂苷 Rg1、Rb1、Rg3 等68单体成分具有抗抑郁活性,可改善 HPA 轴调节紊乱,提高脑源性营养神经因子与单胺神经递质水平9。人参类中药材在 2012 年被国家卫生部批准为药食同源后,在保健食品中的开发应用成为了一大热潮。与园参相比,UPG 中的皂苷种类和含量更为丰富,药用价值更高。但因其生长年限长,对栽培技术,生长环境要求严格,导致 UPG 数量并不多,并且价格较为昂贵而少有研究10。因此本课题采用慢性不可预知温和应激(chronic unpredictable mild str
