探究祛风养心方通过SIRT3-AMPK通路介导自噬抑制Ang诱导H9c2心肌细胞肥大的作用机制。方法 用慢病毒感染H9c2心肌细胞构建SIRT3低表达稳定株。用Ang诱导H9c2心肌细胞肥大模型,制备含药血清。实验组分为4组:10%空白血清(blank)+Lv-NC;Ang+10%空白血清(blank)+Lv-NC;Ang+10%QFYX含药血清+Lv-NC;Ang+10%QFYX含药血清+Lv-shSIRT3。用鬼笔环肽染色评价细胞面积,Western Blot方法检测ANP、BNP、ACT1、LC3-II、Beclin1、P62、P-AMPK、P-mTOR蛋白的表达含量。结果 与blank+Lv-NC组比较,Ang+Lv-NC组H9c2心肌细胞肥大,心肌肥厚基因蛋白的表达上调ANP、BNP(P0.01),ACT1(P0.05),自噬代表基因Beclin1、LC3-II、P62的表达显著上升(P0.01),SIRT3的表达下降(P0.05),P-AMPK下 降(P0.01),P-mTOR上 升(P0.01)。与Ang+Lv-NC组 比 较,Ang+QFYX+Lv-NC组 心 肌肥大不显著,肥厚基因ANP、BNP、ACT1、自噬代表基因Beclin1、LC3-II、P62的表达显著下降ANP、BNP、ACT1、Beclin1(P0.01),LC3-II、P62(P0.05),SIRT3的表达上升(P0.01),P-AMPK上升(P0.01),P-mTOR下降(P0.01),与Ang+QFYX+Lv-NC组比较,Ang+QFYX+Lv-shSIRT3组肥厚基因表达增强ANP、BNP(P0.01),ACT1(P0.05),SIRT3处于低表达水平(P0.01),自噬代表基因的表达升高Beclin1、P62(P0.01),LC3-II(P0.05),P-AMPK下降(P0.01),P-mTOR上升(P0.01)。结论 hypertrophy of H9c2 cardiomyocytes through SIRT3-AMPK pathway.Methods SIRT3 low expression stable strain was constructed by lentivirus infection of H9c2 cells,H9c2 cardiomyocyte hypertrophy model was induced by Ang,prepared drug-containing serum.The experiment was divided into 4 groups:10%blank serum+Lv-NC;Ang+10%blank serum+Lv-NC;Ang+10%QFYX medicated serum+Lv-NC;Ang+10%QFYX medicated serum+Lv-shSIRT3.Phalloidin staining was used to evaluate the cell area,and Western Blot was used to detect the protein expression of ANP,BNP,ACT1,LC3-,Beclin1,P62,P-AMPK and P-mTOR.Results Compared with the 10%blank serum+Lv-NC group,in Ang+Lv-NC group,the expression of cardiac hypertrophy gene protein was up-regulated(ANP,BNP P0.01,ACT1 P0.05),the expression of autophagy representative genes Beclin1,LC3-,P62 were significantly increased(P0.01),the expression of SIRT3 was weaken(P0.05),P-AMPK was decreased(P0.01)accompanied by P-mTOR was increased(P0.01).Compared with Ang+Lv-NC group,myocardial hypertrophy in Ang+QFYX+Lv-NC group was not significantly increased,and the expression of hypertrophic genes ANP,BNP,ACT1,autophagy representative genes Beclin1、LC3-、P62 were significantly decreased(ANP,BNP,ACT1,Beclin1 P0.01,LC3-,P62 P0.05).The expression of SIRT3 was increased(P0.01),with P-AMPK was increased and P-mTOR was decreased(P0.01).Compared with Ang+QFYX+Lv-NC group,SIRT3 in Ang+QFYX+Lv-shSIRT3 group had a low expression level(P0.01),and the expression of autophagy representative genes Beclin1,LC3-and P62 were increased(Beclin1,P62 P0.01,LC3-P0.05),while P-AMPK was decreased and P-mTOR was increased(P0.01).Conclusion The dispelling pathogenic wind and nourishing heart Qufeng Yangxin Decoction can inhibit the hypertrophy of H9c2 cardiomyocytes induced by Ang by improving autophagy and smooth autophagy flow,and this protective effect is achieved by promoting SI
