书书书论著健脾补肾方对辐射损伤小鼠全血细胞及免疫器官的影响研究何东初,肖静静,张梦云,张勇,丁晓娟,伍伟,杨文超(中部战区总医院,湖北 武汉 430070)摘要 目的研究健脾补肾方对辐射损伤小鼠全血细胞减少及免疫器官损伤的干预作用。方法取 90 只昆明种小鼠,随机选择 18 只作为正常组,其余 72 只小鼠采用 6 0 Gy X 射线一次性全身照射。将造模成功小鼠随机分为模型组、健脾补肾方大剂量组、健脾补肾方小剂量组、胸腺肽组,每组 18 只。健脾补肾方大、小剂量组分别给予生药浓度 2 g/mL 和 1 g/mL 健脾补肾方灌胃,胸腺肽组给予胸腺肽肠溶片混悬液 0325 mg/mL 灌胃,正常组和模型组给予生理盐水灌胃,均 2 次/d。每组分别于灌胃第 7,14,30 天各取 6 只小鼠称体重,收集外周血,分离脾脏、胸腺,检测外周血白细胞计数、淋巴细胞计数、红细胞计数、血红蛋白含量、血小板计数及脾脏指数、胸腺指数,HE 染色观察脾脏组织形态学。结果灌胃第 7 天,模型组小鼠外周血白细胞计数、淋巴细胞计数、红细胞计数、血红蛋白含量、血小板计数及胸腺指数、脾脏指数均明显低于正常组(P 均 0 05),脾脏病理改变明显。灌胃第 14 天,模型组小鼠外周血红细胞计数、血红蛋白含量、脾脏指数回升,但仍明显低于正常组(P 均 0 05);白细胞计数、淋巴细胞计数、血小板计数及胸腺指数仍持续下降,脾脏病理改变略减轻。灌胃第 30 天,模型组小鼠外周血白细胞计数、淋巴细胞计数、红细胞计数、血红蛋白含量、血小板计数均回升,但仍明显低于正常组(P 均 0 05);胸腺指数、脾脏指数大致同前,脾脏病理改变略减轻。健脾补肾方小、中剂量组及胸腺肽组不同时间点外周血白细胞计数、淋巴细胞计数、红细胞计数、血红蛋白含量、血小板计数和脾脏指数、胸腺指数均明显高于模型组(P 均 0 05),脾脏病理改变减轻;健脾补肾方小剂量组各指标改善情况优于健脾补肾方大剂量组,促进红细胞计数、血红蛋白含量、血小板计数恢复效果优于胸腺肽组。灌胃第 7 天,健脾补肾方小剂量组红细胞计数、血红蛋白含量恢复正常;灌胃第 14 天,血小板计数、脾脏指数恢复正常;灌胃第 30天,白细胞计数、淋巴细胞计数恢复正常。结论健脾补肾方可以加快辐射损伤小鼠外周血细胞及免疫器官功能的恢复,从而提高机体免疫力,降低感染风险,预防贫血和出血的发生。关键词 健脾补肾方;辐射损伤;全血细胞;免疫器官doi:10 3969/j issn 1008 8849 2022 22 than those of the normal group(all P 0 05),and the pathological changes of spleen were obvious On the 14 th day of gavage,the peripheral blood red bloodcell count,hemoglobin content and spleen index of mice in the model group were increased,but still significantly lowerthan those in the normal group(all P 0 05);The white blood cell count,lymphocyte count,platelet count and thymusindex were continuously decreased,and the pathological changes of spleen were slightly alleviated On the 30th day of ga-vage,the peripheral blood white blood cell count,lymphocyte count,red blood cell count,hemoglobin content and plateletcount of mice in the model group were increased,but they were still significantly lower than those in the normal group(allP 0 05);The thymus index and spleen index are roughly the same as before,and the pathological changes of spleen wereslightly relieved The peripheral blood white blood cell count,lymphocyte count,red blood cell count,hemoglobin content,platelet count,spleen index and thymus index in the small and medium dose groups of Jianpi Bushen decoction and thymo-sin group were significantly higher than those in the model group at each time point(all P 0 05),and the pathologicalchanges of spleen were alleviated;The improvement of each index in the small dose group of Jianpi Bushen decoction wasbetter than that in the large dose group of Jianpi Bushen decoction,and the effect of promoting the recovery of erythrocytecount,hemoglobin content and platelet count was better than that in the thymosin group On the 7 th day of gavage,the redblood cell count and hemoglobin content in the small dose group of Jianpi Bushen decoction returned to normal;The plateletcount and spleen index returned to normal on the 14th day after
