对糖尿病视网膜病变模型大鼠的干预效果郑丽秦学维王利民姚贤凤陈梅伍丹丹(贵州中医药大学第一附属医院眼科,贵州贵阳550001)摘要 目的基于 Wnt 信号通路探究上调 mi-184 对糖尿病视网膜病变模型大鼠的干预效果。方法选取 40 只SD 健康大鼠,10 只作为正常组,剩余 30 只建立糖尿病视网膜病变模型,并随机分为模型组、下调 mi-184 组、上调 mi-184组。对 4 组大鼠分别进行干预,同时对 4 组大鼠视网膜组织进行苏木素-伊红(HE)染色处理。对总胆固醇(TC)、三酰甘油(TG)、空腹血糖水平进行检测对比。采用免疫比浊法、酶联免疫吸附试验分别对谷胱甘肽(GSH)、一氧化氮(NO)、血管内皮生长因子(VEGF)、肿瘤坏死因子(TNF)-水平进行检测。结果与正常组相比,模型组 TC、TG、空腹血糖、VEGF、TNF-、NO水平显著升高(P0.05);与模型组相比,下调 mi-184 组 TC、TG、空腹血糖、VEGF、TNF-、NO 水平显著升高(P0.05);与下调 mi-184 组相比,上调 mi-184 组 TC、TG、空腹血糖、VEGF、TNF-、NO 水平显著降低(P0.05);与正常组相比,模型组 GSH显著下降(P0.05);与模型组相比,下调 mi-184 组 GSH 显著下降(P0.05);与下调 mi-184 组相比,上调 mi-184 组 GSH显著上升(P0.05);与正常组相比,模型组糖原合成激酶(GSK)-3 表达显著升高、-连环蛋白(catenin)、原癌基因(C-Myc)表达显著降低(P0.05);与模型组相比,下调 mi-184 组 GSK-3 表达显著升高、-catenin、C-Myc 表达显著降低(P0.05);与下调 mi-184 组相比,上调 mi-184 组 GSK-3 表达显著降低,-catenin、C-Myc 表达显著升高(P0.05)。结论上调 mi-184 干预糖尿病视网膜病变模型大鼠,对血脂、血糖水平具有调控作用,可改善眼底视网膜环境,有效降低血管通透性,从而达到治疗糖尿病视网膜病变的效果。关键词 Wnt 信号通路;上调
