53现代检验医学杂志第 38 卷第 1 期2023 年 1 月J Mod Lab Med,Vol.38,No.1,Jan.2023白癜风患者血清 LncRNA MALAT1 及 miR-211-5p 表达水平 与临床特征的相关性分析郭雯1，郭建辉1，杜凯晴2，宫克2，申亚娜3（1.河北省沧州中西医结合医院皮肤科，河北沧州 061724；2.河北中医学院研究生学院皮肤科专业，石家庄 050091；3.承德医学院研究生院皮肤科专业，河北承德 067000）摘要：目的探讨长链非编码 RNA 肺腺癌转移相关转录因子 1（long non-coding RNA metastasis associated transcription factor 1 in lung adenocarcinoma，LncRNA MALAT1），微小核糖核酸（micro RNA,miR）-211-5p 表达水平变化与临床特征的相关性。方法选取2022年16月河北省沧州中西医结合医院皮肤科收治的白癜风患者80例为研究对象（白癜风组），同期于该院进行体检的健康者 80 例为对照组。采用实时荧光定量 PCR（quantitative real-time PCR，qRT-PCR）法检测受试者血清中 LncRNA MALAT1，miR-211-5p 表达水平；两组间比较采用独立样本 t 检验；多组间（不同临床分期、临床分型、皮损面积及病程）比较进行单因素方差分析，组间多重比较采用 SNK-q 检验；白癜风患者皮损面积及病程与血清中 LncRNA MALAT1 和 miR-211-5p 表达水平的相关性采用 Pearson 法分析。结果白癜风组患者血清 LncRNA MALAT1（1.900.25）表达水平高于对照组（1.020.13），血清 miR-211-5p（0.530.07）表达水平低于对照组（1.050.10），差异均有统计学意义（t=27.933，38.103，均 P=0.000）。进展期及快速进展期血清 LncRNA MALAT1 水平均高于稳定期（2.290.42,2.470.31 vs 1.540.12），差异有统计学意义（t=15.000，16.766，均 P 0.001）。血清 miR-211-5p 水平均低于稳定期（0.280.09,0.220.06 vs 0.740.11），差异有统计学意义（t=24.748，25.217，均 P 0.001）。寻常型血清 LncRNA MALAT1（2.060.26）表达水平高于节段型（1.500.16），寻常型血清 miR-211-5p（0.390.11）表达水平低于节段型（0.860.21），差异有统计学意义（t=9.768，13.127，均 P 0.001）。泛发型血清 LncRNA MALAT1 相对表达水平高于肢端型、局限型及散发型（2.590.38 vs 2.020.19，1.980.44，1.890.30），差异有统计学意义（t=5.559，6.038，7.375，均 P 0.001）。血清 miR-211-5p 水平低于肢端型、局限型及散发型（0.170.04 vs 0.390.09，0.430.11，0.450.10），差异有统计学意义（t=7.651,9.177，10.520，均 P 0.001）。皮损面积 1%，1%50%和 50%的白癜风患者血清 LncRNA MALAT1 表达水平依次升高（1.690.19,1.920.21,2.560.26），差异有统计学意义（t=6.424,17.408,12.312，均 P 0.001）；皮损面积 1%，1%50%和 50%的白癜风患者血清 miR-211-5p 表达水平依次升高（0.700.09,0.410.10,0.210.03），差异有统计学意义（t=18.972,22.964,9.012，均 P 0.001）。Pearson 相关性分析显示，白癜风患者血清 LncRNA MALAT1 与皮损面积呈正相关（r=0.576，P 0.001），血清 miR-211-5p 与皮损面积呈负相关（r=-0.475，P 0.001）。病程 1 年、1 3 年和 3 年的白癜风患者血清 LncRNA MALAT1 表达水平依次降低（1.650.18,1.880.22,2.480.23），差异有统计学意义（t=5.984，20.581，13.291，均 P 0.001）；病程1 年、13年、3年的白癜风患者血清miR-211-5p表达水平依次降低（0.680.11,0.530.10,0.210.04），差异有统计学意义（t=8.352，24.942，15.170，均 P 0.001）。Pearson 相关性分析表明，血清 LncRNA MALAT1与病程呈正相关（r=0.344，P 0.001），血清 miR-211-5p 与病程呈负相关（r=-0.433，P 0.001）。结论白癜风患者血清中 LncRNA MALAT1 表达上调，miR-211-5p 表达下调，且与皮损面积及病程等临床特征显著相关。关键词：白癜风；长链非编码 RNA 肺腺癌转移相关转录因子 1；微小核糖核酸-211-5p中图分类号：R758.41；R392.11文献标识码：A文章编号:1671-7414（2023）01-053-06doi:10.3969/j.issn.1671-7414.2023.01.011Correlation Analysis of Serum LncRNA MALAT1 and miR-211-5p Expression Levels with Clinical Characteristics in Patients with VitiligoGUO Wen1,GUO Jian-hui1,DU Kai-qing2,GONG Ke2,SHEN Ya-na3(1.Department of Dermatology,Cangzhou Hospital of Integrated TCM-WM Hebei,Hebei Cangzhou 061724，China；基金项目：河北省中医药管理局（2022262）：墨莲祛白汤联合 308nm 准分子光治疗肾虚血瘀型白癜风的临床疗效观察。作者简介：郭雯（1979-），女，硕士研究生，副主任医师，研究方向：中医药防治皮肤病，E-mail：。通讯作者：郭建辉（1982-），男，硕士研究生，副主任中医师，研究方向：银屑病等疑难皮肤病中西医结合治疗，E-mail：。54现代检验医学杂志第 38 卷第 1 期2023 年 1 月J Mod Lab Med,Vol.38,No.1,Jan.20232.Department of Dermatology,Hebei University of Chinese Medicine Graduate School,Shijiazhuang 050091，China；3.Department of Dermatology,Graduate School of Chengde Medical College,Hebei Chengde 067000，China)Abstract:ObjectiveTo investigate the correlation between the expression levels of long non-coding RNA lung adenocarcinoma metastasis-associated transcription factor 1(LncRNA MALAT1)and microRNA(miR)-211-5p and clinical characteristics.MethodsFrom January to June 2022,80 patients with vitiligo who were admitted to the Department of Dermatology,Cangzhou Hospital of Integrated Traditional Chinese and Western Medicine,Hebei Province were taken as the research objects(Vitiligo group).Meantime,80 healthy people who underwent physical examination in the hospital were the control group.The expression levels of LncRNA MALAT1 and miR-211-5p in the serum of subjects were detected by real-time quantitative PCR and independent sample t-test was used for comparison between the two groups.One-way analysis of variance was used for comparison among multiple groups(different clinical stages,clinical types,skin lesion area and disease duration),and SNK-q test was used for multiple comparisons between groups.The correlation between the skin lesion area and the disease course of patients with vitiligo and the expression levels of LncRNA MALAT1 and miR-211-5p in serum was analyzed by Pearson method.ResultsThe expression level of serum LncRNA MALAT1(1.900.25)in vitiligo group was higher than that in control group(1.020.13),and the expression level of serum miR-211-5p(0.530.07)was lower than that in control group(1.050.10),and the differences were statistically significant(t=27.933,38.103,all P=0.000).The serum levels of LncRNA MALAT1 in the progressive stage and rapid progressive stage were higher than those in the stable stage（2.290.42,2.470.31 vs 1.540.12）,and the differences were statistically significant(t=15.000,16.766,all P 0.001).The serum levels of miR-211-5p were lower than those in the stable stage（0.280.09,0.220.06 vs 0.740.11）,and the differences were statistically significant(t=24.748,25.217,all P 0.001).The expression level of LncRNA MALAT1(2.060.26)in serum of vulgaris typ
